Background
TLR5 is a receptor that recognizes flagellin, a protein component of bacterial flagella. Its activation leads to the activation of NF-κB and MAPK pathways, which induce the production of inflammatory cytokines and are essential for sensing flagellated bacteria and initiating the immune response against them.
CD14 is a glycoprotein primarily expressed on the surface of monocytes and macrophages, where it serves as a co-receptor for the recognition of pathogen-associated molecular patterns (PAMPs), particularly lipopolysaccharides (LPS) from Gram-negative bacteria. CD14 exists in two forms: a membrane-bound form and a soluble form. The membrane-bound CD14 facilitates the binding of LPS and other microbial components to the TLR4-MD2 complex, which is critical for initiating innate immune responses. This activation leads to the production of pro-inflammatory cytokines and helps mediate the body's response to infections and inflammation. CD14 also plays a role in modulating immune responses and maintaining immune homeostasis.
