Background
CD80, also known as B7-1, is a co-stimulatory molecule expressed on the surface of antigen-presenting cells (APCs), including dendritic cells, macrophages, and B cells. It plays a critical role in the activation of T cells by providing essential secondary signals necessary for T cell proliferation and survival. CD80 binds to the CD28 receptor on T cells, enhancing their activation when antigen is presented through the T cell receptor (TCR). Additionally, CD80 can interact with CTLA-4, leading to negative signaling that inhibits T cell activation. This dual role makes CD80 essential for regulating the immune response, promoting effective T cell activation while also maintaining tolerance to prevent autoimmune reactions.
CD86, also known as B7-2, is another co-stimulatory molecule found on antigen-presenting cells, such as dendritic cells, macrophages, and B cells. Like CD80, CD86 binds to CD28 on T cells to provide crucial signals that enhance T cell activation, proliferation, and cytokine production. CD86 can also interact with CTLA-4, but it offers a more transient signal compared to CD80. This interaction allows for fine-tuning of T cell responses, acting as a facilitator for activating T cells during the immune response while contributing to regulation and prevention of excessive activation. Together, CD80 and CD86 are fundamental for the initiation and regulation of adaptive immune responses.

Identity testing is required for newly established cell lines. Isoenzyme analysis is used to confirm the identity of the species of a cell line. Alternative methods for identity testing include DNA fingerprinting, STR analysis and karyology.

Frozen cells should be stored in a liquid nitrogen tank (-150°C~-190°C). After thawing, cells should be immediately transferred to the appropriate culture medium and maintained under recommended culture conditions.
Handling Notes
Frozen cells should be thawed immediately upon receipt and grown according to handling procedure to ensure cell viability and proper assay performance.
Note: Do not freeze the cells upon receipt as it may result in irreversible damage to the cell line.
