Introduction of Cytokine Overexpression

In the past few decades, pilot studies have shown that cytokines are different types of proteins that are made by specific immune-related cells, and non-immune cells, such as endothelial cells, and stromal cells. Cytokines play an important role in regulating immune responses against cancer, infections, and other human diseases. It has been divided into many superfamilies, including interferons (INFs), tumor necrosis factors (TNFs), interleukins (ILs), as well as transforming growth factors (TGFs) based on specific structures.

Many reports have revealed that several cytokines secreted by tumor cells can not only create a positive feedback loop in an autocrine manner, stimulate their proliferation, expansion, and drug resistance, but also induce the recruitment, activation, and differentiation of other cells in the tumor microenvironment (TME) in a paracrine manner, like IL-6. IL-8 or VEGF. Moreover, the overexpression of certain cytokines can also stimulate immune cells to secrete more new cytokines. These new cytokines act in an autocrine and paracrine manner, resulting in a chronic inflammatory state that promotes cancer growth.

Recent research has indicated that overexpression of cytokines can induce self-destruction of cancer cells, and It is especially effective against aggressive cancers, such as pancreatic cancer. As a result, understanding this abnormal/overexpression cytokine secretion will enable us to better identify specific disease-causing mutations in cancer cells and thus propose novel tumor immunotherapy approaches.

Our Platform for Overexpressed Cytokine Secretion Kinetics Analysis

In Creative Biolabs, a multitude of different assays have been developed for generating a well-mature and comprehensive cytokine kinetics analysis platform. Our platform can fully evaluate cytokine secretion capacity, especially for overexpression cytokines, and their kinetic properties in various cancer cells. Up to now, a wide variety of advanced technologies, including but not limited to, enzyme-linked immunosorbent assay (ELISA) and flow cytometry (FACS), have been broadly used for cytokine quantification.

Currently, a large number of kinetic parameters can be measured by our platform to offer highly sensitive, accurate, as well as all-around detection of analysts. Our analysis system is suitable for many cytokines types, and performs PK/PD analysis in a variety of samples, for example, blood, cell, serum, or tissue.

In our labs, a typical sandwich ELISA, which uses two highly specific antibodies bound to different epitopes of a protein and simultaneous quantification of all protein targets, has been developed and widely used in overexpressed cytokine secretion kinetics analysis. This assay requires only 25 µL of plasma or serum, or 50 µL of cell culture supernatant, and takes only four-six hours to obtain assay results. In a recent study, The kinetic secretion profiles of IL-2, IL-5, TNFα, IL-1β, IL-6, IL-1α, and IL-8 have been tested in pancreatic cancer cells. The data have suggested that our platform can cater to multiple cytokine assays and can also be customized for new combinations of multiple assays. These data can directly reflect the self-destruction effect of overexpression of different cytokines in cancer immunotherapy.
