Adenosine triphosphate (ATP) assay is another widely used cell viability assay. When the cancer cells undergo apoptosis or necrosis upon treatment with a compound, cellular ATP content decreases and it can be quantified by colorimetric, fluorometric or luminometric way. The latest bioluminescence assays measure cellular ATP content using the enzyme luciferase which is more sensitive and reproducible when determining cell viability compared to conventional assays.
