This publication introduces SDT as a promising, non-invasive cancer treatment that utilizes ultrasound (US) to activate sonosensitizers, generating ROS to damage cancer cells. Unlike photodynamic therapy (PDT), US offers deeper tissue penetration, making SDT favorable for deep-seated tumors.

The paper addresses the challenge of developing innovative sonosensitizers with high efficacy and biocompatibility. The authors synthesized a novel BODIPY-Zn complex, BSS-Zn, incorporating a hydrophilic polyethylene glycol unit, and investigated its potential as a sonosensitizer. BSS-Zn demonstrated enhanced ROS generation upon US irradiation, outperforming a control sensitizer (BSS, a Zn-free analog) and a commercial sonosensitizer (ZnPc), showing increased levels of hydroxyl radicals, singlet oxygen, and superoxide. The study confirmed BSS-Zn's SDT effect on MDA-MB-231 breast cancer cells, identifying specific intracellular ROS types and assessing cytotoxicity. This highlights BSS-Zn's potential as an innovative sonosensitizer, guiding the design of efficient new agents.
