Introduction to Yeast FLP/FRT Site-specific Recombination System

The FLP/FRT system is a typical two-component site-specific recombination system. The full length of the FLP recombinase gene is 1,272 bp, encoding a 48 kD polypeptide monomer protein of 423 amino acid residues. There are two types of mutant recognition sites in the FLP/FRT system, one is an inverted repeat mutation site, and the other is a spacer mutation site. Currently, spacer region mutant FRT sites have been widely used in genetic engineering research in higher eukaryotic model organisms such as drosophila and mice.

As a result, Creative Biolabs has established a yeast FLP/FRT site-specific recombination system, which can achieve precise regulation of tumor-related gene targets in time and space, effectively promoting the rapid development of functional gene research in tumor treatment. Moreover, we have also developed a novel tumor cell lineage tracing technology based on a regular FLP/FRT system to reveal the proliferation, differentiation, and migration processes of tumor cells and their offspring through various labeling methods.

This technique enables the tracking of the growth and spread of thousands of human metastatic cancer cells over six months. In addition, the data obtained from our system can provide information on many other difficult-to-observe aspects of cancer cell biology, like genetic mutations, microenvironmental adaptation, and drug resistance.
