Cardiolipin (CL) was first isolated from the beef heart in the 1940s and is found abundant in the inner mitochondrial membrane, which comprises about 20% of the total lipid composition. CL is a kind of diphosphatidylglycerol lipid characterized by a glycerol backbone connected to two phosphatidyl lipids, so each molecule has four acyl chains and potentially two negative charges. Changes in pH and the presence of divalent cations can lead to a structural change of CL due to its unique composition. For example, it is found that there is a lamellar-to-hexagonal phase transition on CL in the presence of Ca2+ or other divalent cations, which are believed to have connections with membrane fusion. Besides, research has shown that CL also plays a key role in apoptosis, oxidative phosphorylation, and cholesterol translocation, etc.
