Background of Gene Therapeutics

Gene Therapeutics, referred to gene therapy and also called human gene transfer, is a type of treatment that delivers genes (nucleic acids) in patient's cells to treat or prevent disease. Gene therapy actually corrects abnormal genes and alters the expression of the human genes by replacing a faulty gene or adding a new gene, which is expected to improve the ability to fight disease and treat a wide range of incurable diseases, such as genetic diseases, immune diseases, metabolic disorders and cancers. This newly developed therapy dates back to 1990 when Anderson, France, first inserted human DNA directly into the nuclear genome, but has not yet successfully treated human disease.

Lipid-Based Drug Delivery System Applied for Gene Therapeutics

Generally, direct insertion of a gene into a cell is difficult to achieve and may not work. In order to deliver the exogenous genetic material into the cell nucleus, vectors or carriers must be used to deliver the therapeutic gene to the patient's target cells. There are two main types of viral and non-viral vectors, of which viral vectors are commonly used. Viral vectors actually are recombinant viruses that present several risks, such as immune response, infections and non-specific cells targeting. Many scientists believe that lipid-based drug delivery systems, belonging to non-viral vectors are definitely possible to be good carriers for gene delivery. And several liposome-based genes delivery systems have already been evaluated in some cancer treatments in the clinic.

  • Cationic lipid-based delivery systems help negatively charged genes readily diffuse across cell membranes.
  • Encapsulation of lipid carriers can avoid degradation of nuclease
  • Lipid-based genes delivery systems are less likely to trigger an immune response
  • High-capacity of liposomes allows for large pieces of DNA transportation
  • Genes loaded lipid-based delivery system decorated with specific molecules can be targeted to specific cells or tissues
