Magic™ Rabbit TCR Repertoire Profiling

Equipped with world-leading technology platforms and professional scientific staff in the field of immunology and next-generation sequencing (NGS), Creative Biolabs has rich experience in T cell receptor (TCR) repertoire analysis in rabbit. With our powerful Magic™ platform, our seasoned scientists are skilled in sequencing complementarity-determining regions (CDRs) of TCR in the rabbit. Through our high-throughput system, clients could obtain the most comprehensive view of TCR repertoire in the rabbit with reducing project costs and timelines.

Properties of TCR Repertoire in the Rabbit

The rabbit locus spans less than 600 kb and the general genomic organization is highly conserved with respect to other mammalian species. Studies demonstrate that multiple duplication events within a few subgroups have generated the germline repertoire of the rabbit T cell receptor beta variable (TRBV) genes. TRBV in rabbit is substantially larger than those described in humans, mice, and dogs, which suggests that a strong evolutionary pressure has selected the development of a species-specific TRBV repertoire. In rabbits, γ δ T cells compose a sizeable proportion of T cells in the periphery, compared to mice and humans. Greater T-cell receptor gamma (TRG) V-J junctional diversity has been found in tissues from adult compared to newborn rabbits. Rabbits also have a larger germ line-encoded TRG repertoire compared with that of mice and humans. TRGV gene usage is alike in most tissues of rabbits similar to that found in humans but in contrast to that found in mice. Gene conversion happens between TRGV subgroups.

The coding nucleotide sequences of all selected TRBV genes from rabbits, humans, and dogs were combined in the same alignment and an unrooted phylogenetic tree was made using NJ method (Antonacci et al. 2014).

Rabbit TCR Repertoire Analysis Service in Creative Biolabs

Rabbit has provided valuable information for immunological research. For a more comprehensive understanding of TCR repertoire in rabbit, Creative Biolabs has developed a strategy to define the TCR repertoire in the rabbit by analyzing CDRs. Special primers for CDRs of TCR in rabbit have been designed and synthesized for sequencing. We also have developed a powerful bioinformatics tool for analyzing a large number of DNA sequences present in the TCR repertoire.
