The nanodiscs is a non-covalent structure of lipid bilayer and membrane scaffold protein (MSP). MSPs are genetically engineered proteins, which derived from human apolipoprotein A-1 to shield the hydrophobic core of the lipids from the aqueous phase. The size of the nanodiscs was controlled by the length of the MSPs. The diameter of nanodiscs is typically in the order of ~10 nm, but the generation of specific MSP variants allows the formation of smaller (6-7 nm) and larger (16-17 nm) nanodiscs. Lipid-bilayer nanodiscs are the most powerful tool in solubilizing/incorporating membrane proteins in a cell membrane-like environment. This solubilization/incorporation system represents a very suitable option if you need to keep the folded conformation, intact function, and oligomerization of your target proteins all in their native forms.

In nanodiscs assembly, we will first solubilize your membrane proteins using detergents and purify them in a micellar solution. Since different detergents and detergent mixtures may affect the stability, structure, function, and activity of a membrane protein very differently. We can also use our detergent screening platform to find the most suitable detergent or detergent mixture for your membrane proteins. After the solubilization and purification, we will add lipids and the membrane scaffolding protein (MSP) to the micellar solution of membrane proteins followed by removing the detergents via hydrophobic beads binding or dialysis.
