pH-sensitive Lytic Peptide Discovery Service

Lytic peptides with improved stability and acid-activated cell lysis activity have wide biomedical applications, especially for cancers treatment. Creative Biolabs has established an advanced phage display platform to provide a series of services related to Phage Display Library Construction and Phage Display Library Screening. Membranes play an indispensable role in many biological processes, including signaling, membrane trafficking, viral cell entry and propagation. It has been found that the formation of aggregates has great impacts on peptide binding and insertion into the lipid bilayer of the cell membrane. Lytic peptides are a group of cationic membrane-acting peptides, which mainly target the cell membrane and demonstrate strong anti-bacterial and anti-cancer activities. Lytic peptides are characterized by their high arginine and lysine content. These two basic amino acids play a key role in peptide membrane partitioning and contribute greatly to the bioactivity of peptides. Lytic peptides can kill cells by causing cell lysis without the need of getting into cells. The aggregation and dissolution controlled by pH were responsible for the pH-dependent membrane lysis activity of peptides. As solid tumors have consistently low extracellular pH, peptides with acid-activation features showed improved selectivity to cancer cells. As a result, pH-sensitive lytic peptides have excellent activity in both drug-sensitive and drug-resistant cells. An understanding of pH-sensitive lytic behavior will help us to better understand the biological processes, such as endosomal escape of viruses and gene delivery vehicles into the cytoplasm. The unique character of pH-sensitive lytic peptides makes them very attractive for their biomedical applications especially the treatment of cancers, in which drug/antibiotics resistance has occurred.

Based on our advanced phage display platform, Creative Biolabs has developed lots of strategies for isolation of pH-sensitive lytic peptide. By screening our diverse phage display libraries for pH-sensitive lytic peptides with membrane affinity, we have circumvented structural or sequence restrictions used in rational peptide design and overcome the limitations imposed by combinatorial chemistry.
