Prokineticin Receptor Family

Prokineticins are small secreted bioactive peptides, comprising two classes: prokineticin-1 and prokineticin-2. They exert their biological activity by binding to two G-protein-coupled receptors: prokineticin receptor (PKR) 1 and 2. PKR1 and PKR2 belong to the neuropeptide Y (NPY) receptor class, with an overall identity in their amino acid sequences of 85%, differing mainly at the N-terminal. The PKRs are Gq-coupled receptors and promote intracellular Ca2+ mobilization. Besides, they couple to Gi and Gs proteins, indicating the PKRs activate multiple intracellular signal transduction pathways. In humans and rodents, PKRs are distributed in the peripheral tissues and the CNS. This PK/PKR system has been identified and linked to several biological effects as gut motility, neurogenesis, angiogenesis, circadian rhythms, hematopoiesis, and nociception.

Recent studies have revealed that prokineticins and their receptors are associated with the pathologies of various human diseases and genetic syndromes, including abdominal aortic aneurysm, myocardial infarction, heart failure, Kallmann syndrome, and tumorigenesis. Currently, the molecular mechanisms and biological roles of PKRs have not been fully characterized.
