The Properties of Protease

Proteases always perform proteolysis, protein catabolism by hydrolysis of peptide bonds and they have evolved multiple times. Different classes of protease can perform the same reaction by completely different catalytic mechanisms. Proteases are involved in digesting long protein into shorter fragments by cutting the peptide bonds. They exist in all organisms, from prokaryotes to eukaryotes to viruses. These enzymes are involved in a multitude of physiological reactions. Proteases not only can break specific peptide bonds depending on the amino acid sequence of a protein, but also break down a complete peptide to amino acids. The activity can abolish a protein's function or digest it to its principal components. Proteases can be an activation of a function, or it can be a signal in a signaling pathway.

Protease-neutralizing Antibody Discovery Service in Creative Biolabs

Creative Biolabs has developed a novel bacterial genetic strategy to isolate protease-neutralizing antibody from phage display libraries. Our strategy is based on the concerted co-expression of a reporter gene, recombinant protease and fusion-stabilized antibodies (scFv/Fab/VHH) in Escherichia coli. The reporter system has been constructed by inserting a short peptide corresponding to the cleavage site of the protease into a permissive site of the enzyme β-galactosidase. Once isolated, the inhibitors will be validated by an enzyme-linked immunosorbent assay and in vitro catalysis assays.
