TAC, T cell antigen connector, is an alternative receptor designed to redirect T cells in a TCR-dependent, MHC-independent manner. TAC consists of four components, one is an antigen-binding region. This domain is responsible for the binding of tumor antigens, for example, DARPin (anti-HER2) or scFv (anti-CD19). The second part is a target scFv region that is designed for studying various antibodies from different clones. The third part is a T cell receptor recruiting region, such as the CD3 binding domain. The final part is a co-receptor region, include, CD4, CD8. TAC receptor can trigger aggregation of the native TCR following binding of tumor antigens by co-opting the native TCR via the CD3 binding domain. In these three TAC domains, each domain can be designed to use a variety of proteins, allowing the TAC to be modified to best respond to a large number of different antigens.
