DSPE-PEG-CHEMS is a pH-sensitive component for liposomal formulations. It is composed of a DSPE lipid covalently linked to a PEG chain that is terminated with Cholesteryl Hemisuccinate (CHEMS). CHEMS is a cholesterol derivative that confers pH-dependent stability to lipid bilayers. It acts as an anionizable lipid, stabilizing membranes at neutral pH but becoming protonated and disrupting membrane integrity in acidic environments (e.g., endosomes), facilitating content release and endosomal escape.
