Targeting Delivery for Gene Editing

Gene editing technology represents the highest precision in biomedicine, offering the potential for curative therapies. However, its clinical safety and effectiveness depend entirely on whether the editing tool can reach the target gene locus in the most efficient and precise way. Creative Biolabs' targeted delivery technology focuses on overcoming the challenges of efficiency and safety in gene editing tool delivery, helping you achieve "precise rewriting" of the genome.

What is Targeted Delivery?

Targeted delivery here specifically refers to the safe, efficient, and cell-specific delivery of the core components of a gene editing tool—such as the Ribonucleoprotein (RNP) complex—into the nucleus of the target cell. This process requires navigating complex biological barriers, including endosomal entrapment, before cargo release. Ensuring the instantaneous presence and effectiveness of the editing tool in the target cell minimizes off-target editing and systemic immune responses, establishing the core strategy for achieving safe and therapeutic gene modification.

Applications for Gene Editing

Precision Treatment of Genetic Diseases

Delivering high-fidelity Cas9 RNP complexes via optimized nanovesicles for in vivo or in vitro knockout or precise correction of disease-causing genes. This includes successful non-viral editing of hepatic genes, such as permanent PCSK9 correction for hypercholesterolemia.

Universal Cell Therapy Manufacturing

Utilizing advanced non-viral vector solutions to deliver gene editing tools for in vitro knockout of endogenous TCR or HLA genes, accelerating the development of universal products such as CAR-T/TCR-T with enhanced scalability.

Base & Prime Editing

Facilitating the delivery of complex, large-sized editing proteins and gRNAs. This enables more precise single-base mutation repair, advancing therapies that require highly localized, transient editing and avoiding unintended double-strand breaks.

Delivery Challenges of Gene Editing

Off-Target Editing & Safety: Prolonged intracellular residence time (e.g., from long-term expression of delivered mRNA) or non-specific delivery of editing tools increases the risk of off-target editing and potential genotoxicity or tumorigenesis.

Efficient Delivery of Large-Molecular RNPs: Cas9 nuclease is a large protein molecule. Efficiently and intactly delivering it to the cell nucleus after forming an RNP complex with gRNA presents a significant delivery challenge, particularly the critical hurdle of endosomal escape.

In vivo Targeting & Immunogenicity: Viral vectors (such as AAV) pose risks of immunogenicity and sustained expression when delivering the Cas9 gene in vivo. Conversely, the in vivo efficiency and specific tissue tropism of untargeted non-viral vectors (LNPs) still require sophisticated surface optimization.

What We Can Offer

We offer professional targeted delivery technologies via our proprietary platform, focusing on solving the challenges of efficiency, safety, and precision in gene editing:

Ultra-Efficient RNP Delivery Platform

Optimized LNP/nanogel vectors, specifically designed for Cas9 RNPs, utilize stimuli-responsive cationic lipids to achieve highly efficient, low-toxicity nuclear delivery by promoting endosomal escape, ensuring the highest instantaneous editing efficiency.

In vitro Cell Engineering Optimization

Providing customized electroporation or non-viral vector solutions to optimize RNP delivery for challenging cells like T cells, NK cells, and hematopoietic stem cells, facilitating the universalization of cell therapy.

In vivo Targeted Delivery Innovation

Optimized vector surface modification, including the use of aptamers for receptor-mediated endocytosis, enables precise targeting of non-hepatic organs such as the nervous system and muscles, expanding the application scope of in vivo gene editing.

Editing Efficiency & Safety Assessment

Provides full-process support, including RNP activity detection, off-target analysis, and editing efficiency verification, ensuring high precision and low toxicity in editing.

Why Choose Us?
