Baculovirus-Insect Expression System

Baculovirus Expression Vector System (BEVS) is becoming a mainstream method for vaccine production these years. Besides, a number of viral vaccines based on BEVS are under development.

How BEVS Platform Works to Produce Vaccines?

BEVS utilizes recombinant baculovirus to specifically infect insect cells for antigen production. To initiate the process, a recombinant baculovirus that consists of genes of interest is constructed. In this step, foreign gene of antigen is inserted into a transfer plasmid, commonly behind the strong promoter to attain high-level expression of desired protein. The inserted gene is flanked by homologous sequence of baculovirus. Next, insect cells are co-transfected with recombinant transfer plasmid and linearized DNA of baculovirus. During this stage, homologous recombination takes place and a recombinant baculovirus which includes genes of desired antigen is generated. Then, such baculovirus is plated and purified through plaques trials. The selected plaque is later passaged for several generations in insect cell to stabilize virus and establish a working virus bank (WVB) for further production. At last, WVB is used to infect insect cells at their logarithmic growth phase cultured in bioreactors or in flasks for large-scale production or for small-scale production respectively. After the recombinant baculovirus infects insect cells for 48-96 hours, the target protein is expressed in the cell or secreted into the culture medium. Kinds of downstream processing are applied to isolate and purify the protein.

For laboratory research, quick and easy manipulations for construct of recombinant baculovirus is required. Commercialized kits such as Bac-to-Bac and other similar kits enable researchers to complete the construction of recombinant baculovirus in E. coli, simplifying and improving the process and efficiency of small-scale studies. However, the shortcoming of such a kit is that the recombinant baculovirus constructed by this method is genetically unstable, and its gene is easy to be lost in the process of virus passage.
