Arenavirus Based Vaccines

Traditional vaccines are generally attenuated or inactivated pathogens, primarily for infectious diseases. In recent years, viruses have received increasing attention as vaccine vectors because they are capable of inducing strong immune responses without the aid of adjuvants. Currently used viral vectors include adenoviruses, poxviruses, and alphaviruses, but these live viral vectors often involve pathogenicity, so the pursuit to find safer immunogenic vectors still requires effort.

Arenavirus is an enveloped double-segment (L segment and S segment) negative-strand RNA virus that encodes four proteins: RNA-dependent RNA polymerase L, ring-finger motif containing Z protein, nucleoprotein NP, and the glycoprotein GPC. Arenaviruses can target antigen-presenting cells such as dendritic cells and macrophages early in infection, and they have multiple strategies to evade the host's natural immune recognition. There are at least 24 currently known species belong to arenavirus family, many of which are believed with potential in prophylactic and therapeutic applications. Lymphocytic choriomeningitis virus (LCMV) is one of them which has been used over ten years in research and has been identified as a useful tool as vaccine vector as it has been proved to be able to elicit the most potent immune responses. Moreover, LCMV with genetically engineered tri-segmented genome has been showed comparable expression levels of foreign genes of interest to that of indigenous viral gene products. Another arenavirus- recombinant tri-segmented Pichinde virus (PICV), is more advantageous in avoiding pre-existing anti-vector immunity (a troublesome issue which often needs extra efforts when using viral vectors as vaccines) as it is only found in Colombia, South America.
