FSPs and FSPs Based Cancer Vaccine

Microsatellites, also known as simple repeats, are repeats of small fragments of nucleotides present in the genome. The repeating units are generally composed of 1-6 nucleotides, and the copy number of the repeating unit is different, so the length of the microsatellite is also not exactly. Microsatellite sequences are highly conserved and stable in inheritance and belong to non-transcribed and constitutive genes, so they can be classified into a class of highly polymorphic genetic markers for individual identification and human genetic analysis. During the process of DNA replication, errors such as base mismatches may occur. These errors are accumulated and passed down from generation to generation, eventually leading to genetic mutations leading to cell cancerization. Mismatch repair systems in the human body correct errors in DNA replication. For example, the base mismatch repair gene (MMR) recognizes and fixes errors in the next generation of cells. Once the MMR gene mutated, some simple repeat insertions or deletions occur during DNA replication, ie, microsatellite instability (MSI). MSI indicates that the size of the structural allele of the tumor tissue and its corresponding non-tumor tissue has changed or that there are insertions or deletions of repeat sequences.

Frameshift peptides (FSPs) are expressed in this case. FSPs are neo-epitopes that elicit effective anti-tumor immune responses, and they have been shown in experiments that FSPs can also induce strong immune responses. Therefore, FSPs with such high immunogenicity may also elicit high level of immune responses in cancer patients. The current study found that endometrial cancer, intestinal cancer, upper urinary tract cancer, colorectal cancer, and pancreatic cancer are all related to MSI to varying degrees. Therefore, this is the data basis for developing cancer vaccines using FSPs as targets. Here, we have developed FSPs-based cancer vaccines by using synthetic peptide technology based on the specific expression of FSPs in cancer and its strong immunogenicity. Preclinical evaluations show that the vaccine can effectively stimulate humoral and cellular immune responses against cancer and can be used as a promising cancer vaccine.
