Modular NeoVax Platform

Vaccine development utilizes the self-nonself discrimination mechanism of the immune system. Different from the infectious diseases causing pathogens, tumor cells derive from normal cells often do not express any differentiating molecular markers from normal cells. However, most tumor cells can express different types of tumor antigens, one is tumor-associated antigen with low expression level in normal tissues and high expression level in tumor tissues; the other is the tumor-specific neoantigen caused by tumor genome mutation, which is not present in normal tissues and thus avoids accidental injury to normal tissues. NeoVax takes advantage of this property of tumor neoantigens.

NeoVax modularly includes three main functional components: a targeting module, a dimerization module, and a tumor neoantigen module. The targeting module enables the vaccine to target and bind to antigen-presenting cells (APCs), which are then internalized into APCs and mediate the maturation of APCs to initiate immune responses. The benefit of this design is that it increases the efficiency of internalized neoantigens’ presentation to helper T cells and cytotoxic T cells and increases the intensity of the immune response. In addition, the modular form also makes it easier to design individualized vaccines and to modify them.
