TCR-T Therapy

TCR-T therapy is a type of adoptive cell transfer therapy (ACT), which improves the recognition of tumor-associated antigens by T cell receptors (TCRs) through transducing chimeric antigen receptors or TCRα/β heterodimers, and enhances affinity of TCRs to tumor antigens and strengthens T cells’ lethality to tumor cells. By infusion of genetically modified T cells that can efficiently identify tumor-specific targets, TCR-T therapy gives the immune system new, unnatural immune activity. In addition to being able to kill tumors as quickly as chemotherapy and targeted therapy, TCR-T therapy can avoid the delayed effects of cancer vaccines and immune checkpoint therapies.

NeoTCR-T Platform - Confer T Cells Ability to Specifically Target Tumor Cells

The general steps of TCR-T therapy are: first identify one or more tumor antigens as therapeutic targets, and then obtain TCR sequences that specifically recognize tumor antigens; then use genetic engineering techniques to introduce the antigen-specific TCR gene sequence in patient-derived T cells, TCR-T cells that specifically recognize tumor antigens are thus obtained. These TCR-T cells are expanded in vitro and then reinfused to patients to kill tumor cells. At present, the clinical effectiveness of TCR-T therapy is still relatively low, and the bottlenecks that limit its effectiveness are mainly the search for effective high-affinity tumor target antigen TCR and the improvement of TCR-T construction and preparation efficiency.
