Poliovirus Vaccines - Creative Biolabs

Poliovirus of the family Picornaviridae is a small, non-enveloped single-stranded positive-sense RNA virus of about 30 nm in diameter and spreads from person to person by the oral route. It is the etiologic agent of the paralytic disease which is called poliomyelitis. The virus is a serious disease affecting a person’s brain and spinal cord and lives in an infected person's throat and intestines. The poliovirus infection can result in a life-threatening muscular paralysis of the body. Most people infected with the poliovirus will not have any visible symptoms. About 25% of infected people will have flu-like symptoms that may include: fever, fatigue, nausea, vomiting, headache, and stomach pain.

There have two types of vaccine against poliovirus which are oral poliovirus vaccine (OPV) and inactivated poliovirus vaccine (IPV), respectively.

Oral Poliovirus Vaccine

OPV consists of three live attenuated poliovirus strains and induces both a systemic and mucosal immune response. OPV protects the individual against polio paralysis by preventing the spread of poliovirus to the nervous system due to it can produce antibodies in the blood to all three types of poliovirus. OPV strains also produce a local immune response in the primary site for poliovirus multiplication which is lining of the intestines. The advantages of OPV vaccine are easier to administrate, superior mucosal immunity, secondary spread to contacts, ease of administration, lower price and had a herd effect, inducing long-lasting protective systemic, humoral and cellular immunity resistance to PV infection. The data shows that one dose of OPV produces immunity against all three PV serotypes in approximately 50% and three doses produce immunity in more than 95% of recipients.

Inactivated Poliovirus Vaccine

IPV is developed from wild-type poliovirus strains of each serotype that have been inactivated with formalin. As an injectable vaccine, it can be administered alone or in combination with other vaccines. Children should get 4 doses of IPV at these ages: 2 months, 4 months, 6-18 months, and a booster dose at 4-6 years. IPV can reduce the predominant mode of transmission even in fecal-oral spread places. Generally, three spaced doses are administered to produce adequate levels of seroconversion resulting in protection against paralytic poliomyelitis. IPV is a safe vaccine for routine infant and childhood immunization because it does not cause vaccine-associated paralytic poliomyelitis and the adverse events following administration of IPV are very mild and transient.
