Clostridium family are heterogeneous genera of saprophytic, gram-positive, spore-forming anaerobes comprised of at least 209 species and 5 subspecies. Although primarily found in soil, Clostridia are commonly found among the gut flora. Pathogenic species such as Clostridium botulinum, C. perfringens, C. difficile and C. tetani, each of them can produce one or more unique exotoxins that sicken the host and can eventually result in death.

Vaccines for Clostridium tetani

Tetanus is a potentially fatal disease of the nervous system caused by Clostridium tetani (an anaerobic bacterium found in the soil). This disease is characterized by muscle stiffness, spasms, and autonomic dysfunction. Tetanus toxin produced by Clostridium tetani (C. tetani) blocks inhibitory neurotransmitters in the central nervous system and causes the muscular rigidity and spasms typical of generalized tetanus. In addition to strengthening public safety and health management, the use of tetanus vaccines has greatly reduced the incidence of the disease and is considered to be one of the most effective measures to prevent tetanus.

Vaccines for Clostridium perfringens

Clostridium perfringens is a Gram-positive, spore-forming anaerobic bacterium which can cause foodborne illness in humans and is responsible for a number of severe infections, including necrotic enteritis in animals, most notably in pigs and poultry. As more and more antibiotics are banned from use in animal feed, alternative strategies to prevent necrotic enteritis are being actively sought, including vaccination against Clostridium perfringens bacteria and vaccination against the toxins they produce, which are responsible for much of the damage observed in infected animals. The conventional vaccine production systems need time-consuming detoxification and difficult quality control steps; therefore, it is imperative to develop a new, non-toxic and efficient new Clostridium perfringens bacterial vaccine.

Vaccines for Clostridium botulinum

Botox poisoning is a severely fatal disease characterized by flaccid paralysis due to the inhibition of acetylcholine release at the neuromuscular junction caused by the botulinum neurotoxins (BoNTs) produced by Clostridium botulinum (C. botulinum). Although post-exposure delivery of antitoxins is effective for the treatment of rare cases of life-threatening botulism, such therapies would be inadequate to prevent intoxication of large populations threatened by the dissemination of botulinum toxins during a bioterrorism event. Therefore, it is very important to develop new therapies and vaccination strategies to prevent poisoning. There are a number of botulinum toxin vaccines currently under development that is expected to be an alternative to specifically targeting botulinum antigens.

Vaccines for Clostridium difficile

Clostridioides difficile, an important human gastrointestinal tract pathogen, causes antibiotic-associated diarrhea in humans, and its symptoms are mainly attributed to two C. difficile toxins, TcdA and TcdB, which have been identified as key virulence factors and also been targeted in vaccine development strategies. A great deal of effort has been put into developing vaccines that target both toxins through the parenteral immunization pathway. C. difficile is an intestinal pathogen, and mucosal/oral immunity is particularly useful for protecting hosts from Clostridium difficile infection, considering that the gut is a major site of disease onset and development.
