Idiotype (ID) is an antigen-specific marker unique to each antibody molecule, and its epitope is called an idiotope. Anti-idiotype antibodies (anti-ID) refer to antibodies that can specifically bind to the idiotope located in the variable region of the antibody to be tested. Idiotype and anti-idiotype antibodies can form a complex network and play an important role in immune regulation.
Because it mimics the antigenic determinants of natural antigens, has the immunogenicity and reactogenicity of natural antigens but lacks other properties, it has advantages in vaccination and immunological diagnosis of epidemics of viral infectious diseases.
According to the different binding modes, anti-idiotypic antibodies can be divided into antigen-blocking type, antigen-non-blocking type and drug-target complex type.

Antigen-blocking anti-idiotypic antibodies recognize the variable region of the antibody drug and the complementary site of antigen binding, can block the binding of antigen and antibody drug, and can be used to detect free antibody drugs.
Creative Biolabs utilizes target antibodies in scFv forms or using peptides designed according to the sequences of the variable domains as targets to isolate anti-idiotypic antibodies from immunized phage display library. If variable regions of both the heavy- and light-chains contribute idiotypic determinants, the idiotype is likely to be a conformational determinant, then scFv form will be used. On the other hand, individual idiotypes may be located either in the heavy- or light-chains and contain the primary amino acid sequences of the variable regions. These idiotypes are more likely to be sequence-dependent, and linear antigenic determinants. In this situation peptides will be utilized as target proteins. Anti-ID antibodies can then be isolated by immunized phage display library technology.

Following booster immunization with tetanus toxoid, researchers investigated the presence of auto-anti-idiotypic antibodies in the serum of two individuals. IgG samples were analyzed for their ability to bind specifically to radiolabeled anti-tetanus toxoid IgG F(ab')2 and to inhibit 125I-radiolabeled tetanus toxoid binding to peak IgG. Auto-anti-idiotypic IgG was detected as early as two weeks post-immunization and persisted throughout the four-month study. Evidence for the modulation of idiotypic expression on anti-tetanus toxoid IgG F(ab')2 was gathered through inhibition assays. Diminished expression of idiotypic determinants was concurrent with the emergence of auto-anti-idiotypic antibodies, suggesting a role in modulating idiotypic expression post-immunization.
