Introduction to DDB1

Damage-specific DNA binding protein 1 (DDB1) is a conserved protein that consists of three seven-bladed β-propeller (BP) domains, BPA, BPB and BPC, and a C-terminal helical domain (CTD). It is a component of the damaged DNA binding protein (DDB) complex and was first discovered in the nucleotide excision repair pathway (NER). In humans, the DDB complex is part of a higher-order DDB1-DDB2-Cul4A complex, wherein DDB1 functions as an adaptor bridging DDB2 to the ubiquitin E3 ligase Cul4A during global genomic nucleotide excision repair (GGR). Also, DDB1 can interact with several partner proteins to regulate the physiological processes which have great relevance to the development of human cancer, such as cell cycle and UV-damaged gene transcription.

DDB1 Marker for Follicular Adenoma

Follicular adenoma, a type of benign and encapsulated nodule in the thyroid gland, has an increased potential of progressing to follicular carcinoma. Therefore, it is necessary to develop an efficient and novel biomarker to monitor the state and progress of follicular adenoma in clinical management to prevent its worsening from follicular carcinoma. Studies have shown that DDB1 has the potential to act as a biomarker for in vitro diagnosis of follicular adenoma as the blood level of DDB1 in follicular adenoma tissue is higher than that in normal tissue.
