BsIgG is one of the widely used BsAb formats. BsIgGs remain the general size and structure of a conventional IgG, bispecific, and bivalent (1+1 antigen binding). They have the most similar biophysical properties, such as serum high stability, half-life and tissue penetration, to the parental mono-sepcific IgGs. The Fc portion can also be engineered to encreace or decrease the Fc-mediated functions, including ADCC, ADCP, CDC, cytokine storm, half-life, etc.
Correct heterodimerization of the two half of the parenatl IgGs is a great challenge in the generation of BsIgGs. Now there have been some strategies that have been successfully used to over the heavy chain-heavy chain mispairing problem, such as Knob-into-holes (KIH), DuoBody, Charge pair, SEEDbody. The technologies including domain crossover, common light chain can force heavy chain-light chain correct pairing.
