Background

IGF-1R, activated by the ligands insulin-like growth factor-1 (IGF-1) or insulin-like growth factor-2 (IGF-2), belongs to the insulin receptor (IR) family. The expression and activity of IGF-1R increase in many tumor types, including rhabdomyosarcoma and ovarian cancer. Functions of IGF-1R include but are not limited to:

IGF-1R Signaling Pathway

IGF-1R is involved in multiple signaling pathways via IRS and PI3K.

• Mediates apoptosis-inhibiting signals, and promotes protein synthesis and cell metabolism by downstream MTOR kinase signaling activates the PI3K/AKT signaling pathway.
• Activates the Grb2/RAS/RAF/MAPK signaling pathway to transduce cell proliferation and growth signals.
• Associated with metastasis and invasion of cancer cells, processes mediated by many signal transduction proteins affect invasiveness.
• Interacted with steroid hormones and other RTKs.
• Intervened by RTK heterodimers, including the INSR hybrid receptor, and IGF-1R/INSR.
• Kinase activity within cells is regulated by notch and apoptosis inducing factor-1.
