Isobaric Amino Acids Discrimination

Monoclonal antibody sequencing plays a significant role in enhancing antibody affinity and humanization by obtaining the first-order antibody sequence. Creative Biolabs provides de novo antibody sequencing services highlighted with precise discrimination of isobaric amino acids.

Isobaric Amino Acids Discrimination

Isobaric amino acids refer to amino acids with the same mass-to-charge ratio (m/z) in mass spectrometry, making them difficult to be distinguished on the mass spectrum, which can impact the accuracy of antibody sequence determination. Analyzing mass spectrometric data containing isobaric amino acids may require more sophisticated algorithms and data processing methods.

Our DASS (Database Assisted Shotgun Sequencing) technology platform, in contrast to other MS-based approaches, is capable of discriminating the majority of isobaric amino acid combinations, including but not limited to:

  • W can be distinguished from GE, AD, and SV.
  • K and Q can be distinct from one another.
  • R can be distinguished from GV.
  • N can be distinguished from GG. Bond breakage between GG is guaranteed by our DASS technology. Additionally, we ensure that asparagine is not GG by using our exclusive derivatization method.
  • Q can be distinguished from GA. The link breaking between GA is ensured by our DASS technology. Additionally, we ensure that glutamine is not GG by using our specialized derivatization method.

Why Choose Us

Our Sequencing

  • Equipped with state-of-the-art sequencers
  • Match 3800 peptides simultaneously
  • Breaking through the low-resolution limitation of traditional sequencing

Traditional Sequencing

  • Lower precision mass spectrometer
  • Need to utilize part of the database information, unable to achieve full sequence coverage
  • Difficult to distinguish between amino acids with close molecular weight (e.g. the difference in molecular weight between glutamine and lysine is only 0.03D)

Direct sequencing analysis of existing antibody products has the potential to accelerate the development of antibody medications, diagnostic tools, and other antibody-based products. Precise antibody sequencing also allows for the optimization of antibody properties, such as affinity and specificity, leading to the development of antibodies with enhanced therapeutic and diagnostic potential. Creative Biolabs features highly efficient and more accurate de novo antibody sequencing technology that can greatly improve the precision of discriminating isobaric amino acids.
