The strategy of single B cell screening to avoid both hybridoma fusion and the combinatorial display has become an important technology for the effective collection of natural antibody repertoire of immunized animals and humans. By increasing the availability of these repertoires, single B cell antibody technology may lead to the development of novel monoclonal antibody (mAb) therapies. This new antibody production technology increases the number of antibodies for different applications, so an efficient production system is urgently needed. At present, more and more recombinant expression systems have been developed from Gram-negative and positive bacteria, yeasts, filamentous fungi, insect cell lines, and mammalian cells to transgenic animals and plants.

Due to the short doubling time, bacteria can be used as a rapid and easy system for expressing antibodies. The advantage is that the culturing cost is low, and the method suitable for large-scale production is very simple. The most widely used prokaryotic host system is Escherichia coli (E. coli), since of which genetics, genome sequence, and physiology have been fully studied, making genetic manipulation easy.
Eukaryotic cells have developed an advanced folding, post-translational, and secretion device that, compared to bacteria, can enhance the secretion of antibodies, including intact immunoglobulins.
Yeast expression system, as a late-generation foreign protein expression system, has the advantages of both prokaryotic and eukaryotic expression systems, and is being increasingly used in the field of genetic engineering. In addition to the earliest application of Saccharomyces cerevisiae, we also provide the most widely used methanol yeast expression system.
Insect cell expression system, especially baculovirus expression system, is widely used in various fields of genetic engineering as yeast expression system because of its safe operation and high expression. The commonly used lines are Sf-9 and Sf-21 of Spodoptera frugiperda, DS2 of Drosophila melanogaster, or High Five cells of Trichopulsia ni.
In terms of activity, the exogenous proteins produced by mammalian post-translational reprocessing are much better than prokaryotic expression systems and eukaryotic expression systems such as yeast and insect cells, and more close to natural proteins. The host cells commonly used in this expression system include CHO and HEK293. Different host cells have different effects on protein expression levels and protein glycosylation. Therefore, the selection of host cells should be based on the specific situation.
More and more attention has been paid to the development of transgenic plants expressing recombinant antibodies, especially in the case of large-scale expression. Compared with other systems (such as mammalian cell culture), the scale-up of this production system is easier to achieve, but the scale-up of the fermentation process leads to an increase in production cost.
