Serial limiting dilution has been used by laboratories and companies all over the world to isolate single cells in recent decades, especially since the application of pipetting robots greatly reduces the workload of personnel. In principle, due to the statistical distribution of the cells in the suspension, the number of cells in the high-dilution sample will be as low as one single cell in a small volume when the suspension is serially diluted into small volumes. Single-cell isolation carried out by manually limiting dilution or pipetting robots is indeed simple but not efficient. Since the probability to obtain a certain number of cells or a single cell in an aliquot is of Poisson’s distribution, adequate dilution of the cell sample is necessary to obtain a sufficiently high probability of the presence of a single cell while minimizing the probability of multiple cells.
This limiting dilution once has been widely applied for the antibody production of monoclonal cell cultures and microbial samples. What is noteworthy is that cell isolation by limiting dilution is random sorting and prone to a high false-positive rate, and not suitable for cell isolation from complex or rare samples.

The single B cell antibody technology has greatly shortened the lead time of therapeutic antibody discovery and production. Before target antibody expression, it is necessary to isolate antibody-secreting B cells to obtain the gene information of the target antibody. For those hosts with high levels of antibodies in the circulating, isolating single antibody-secreting B cells from blood samples can be implemented by this reproducible and relatively cost-efficient limiting dilution. However, to isolate antibody-secreting B cells, it is recommended to combine with upstream sorting (such as B cell labeling) or enrichment techniques. For high-throughput B cell isolation or those individuals with low antibody titer, Creative Biolabs also provides antigen-specific B cell sorting based on a variety of sorting technologies.
