Introduction to Micromanipulation Technique

Micromanipulation technique, also known as manual cell picking, was used to be one of the most widely used cell isolation approaches that artificially isolate target cells using a micromanipulator on a microscopic level. Usually, micromanipulators for cell isolation consist of an inverted microscope and a tiny pipette that can be moved by motorized mechanical stages. Each individual cell in the cell suspension can be observed and photographed under the microscope. After observation, select a target cell and move the micropipette closer, the cells are aspirated by pumping the micropipette.

Cell isolation by micromanipulation technique is a convenient and efficient method for randomly isolating single cells. Distinct from laser capture microdissection that isolates single cells from solid samples, micromanipulation enables the controlled isolation of target living cells from suspension, which is especially suitable for the cell separation of blood samples, and even allows for isolation of prokaryotic cells.

B Cell Random Isolation by Micromanipulation for Antibody Development

When comes to random isolation of single B cell from blood samples of vaccinated individuals, micromanipulation is usually recommended due to its simple operation and no need for special equipment. As early as 1993, scientists isolated B cells by micromanipulation to trace the B cell development in the human germinal center. However, the micromanipulation technique is limited when it comes to high-throughput B cell isolation since this B cell isolation is inefficient and usually requires highly skilled professionals to perform. The harvested B cell selected by the micromanipulator can be cultured to obtain a sufficient number of cells for subsequent transcripts extraction or obtain the target gene information directly by single B cell RNA sequencing.
