The vector of anti-CD70 KIR chimeric antigen receptor (CAR) is constructed for the engineering of T cells to target Mouse CD70. The T cells are genetically modified through transduction with a lentiviral vector expressing scFv (XW-212) of anti-CD70 antibody linked to KIR2DS2 TM & ICD and DAP12. The vector product was designed for the discovery and development of cellular therapy against Colorectal cancer. This KIR signaling is designed to prolong KIR-CAR T cells' efficacy period by coupling the receptor chains during activation and decoupling them during rest, avoiding T cell exhaustion even in challenging solid-tumor environments.
