| Introduction | ABL1 is a non-receptor tyrosine kinase containing an SH3 domain, an SH2 domain, and a C-terminal kinase domain. It localizes to both the cytoplasm and nucleus, where it regulates cell proliferation, cytoskeletal dynamics, and DNA damage responses. |
| Function | ABL1 drives oncogenic signaling through constitutive kinase activity in certain fusion variants. Its well-characterized ATP-binding pocket can be selectively engaged by bifunctional chemical agents, which redirect it toward proteasome-dependent degradation and thereby abrogate aberrant downstream proliferative signaling. |
