Introduction ABL1 is a non-receptor tyrosine kinase containing an SH3 domain, an SH2 domain, and a C-terminal kinase domain. It localizes to both the cytoplasm and nucleus, where it regulates cell proliferation, cytoskeletal dynamics, and DNA damage responses.
Function ABL1 drives oncogenic signaling through constitutive kinase activity in certain fusion variants. Its well-characterized ATP-binding pocket can be selectively engaged by bifunctional chemical agents, which redirect it toward proteasome-dependent degradation and thereby abrogate aberrant downstream proliferative signaling.
