Introduction EED is a core component of the Polycomb repressive complex 2 (PRC2), containing seven WD40 repeats that form a beta-propeller scaffold. It binds to histone H3 methylated at lysine 27 and is essential for PRC2-mediated gene silencing.
Function EED is a structural scaffold protein that allosterically activates the EZH2 methyltransferase within PRC2, driving H3K27 trimethylation and epigenetic gene silencing. Its histone-binding pocket can be targeted by bifunctional molecules to induce proteasomal degradation, disrupting PRC2 assembly and reactivating silenced tumor suppressor genes.
