Introduction Kelch-like ECH-associated protein 1 (KEAP1) is a substrate adaptor for the CUL3-RBX1 E3 ligase complex. It contains a BTB domain for CUL3 binding and a double-glycine repeat (Kelch) domain for substrate recognition.
Function KEAP1 is a specialized BTB-Kelch substrate adaptor within the CRL3 complex that natively regulates oxidative stress responses. In targeted destruction systems, hijacking KEAP1 through its Kelch domain allows bifunctional molecules to tap into a highly responsive, tissue-specific degradation machinery, expanding the available E3 repertoire beyond ubiquitously expressed ligases.
