Introduction Von Hippel-Lindau tumor suppressor (VHL) is the substrate recognition subunit of a CUL2-based E3 ubiquitin ligase complex. It contains an elongin binding domain and a β-domain that recognizes hydroxylated proline residues.
Function As a pioneer E3 ligase in proximity-induced degradation, VHL (Von Hippel-Lindau) forms the core of the CRL2-VHL complex. It is highly valued for its predictable structural rigidity, low active-site flexibility, and robust expression across most tissue types. VHL allows bifunctional degraders to form highly stable ternary complexes, driving efficient neo-substrate polyubiquitination and proteasomal clearing.
