  • How does this work
    The calcium phosphate transfection method used to introduce DNA into mammalian cells is based on the formation of calcium phosphate-DNA precipitates. Calcium phosphate promotes the binding of DNA to the cell surface. The DNA then enters the cell through endocytosis. This method was originally developed by Graham and van der Ebb, and was later modified by Wigler. This procedure is commonly used to transfect a variety of cell types for transient expression or to produce stable transformants. The DNA is directly mixed with concentrated CaCl2 solution and then added dropwise to phosphate buffer to form a fine precipitate. The addition of DNA-CaCl2 solution while aerating the phosphate buffer helps to ensure that the precipitate formed is as fine as possible, which is important because clustered DNA will not effectively adhere or enter cells.
