Antibody/Peptide Discovery Overview

  • Antibody/peptide screening. Various screening methods, such as immunoscreening, affinity screening, etc., are applied to screen candidate molecules with specific functions and activities from a large number of antibodies/peptides.
  • Antibody/peptide optimization. The screened antibodies/peptides are optimized to improve their properties such as affinity, stability, solubility, etc., as well as to reduce potential immunogenicity.
  • Pharmacokinetic studies. The absorption, distribution, metabolism and excretion processes of the antibody/peptide are studied in vivo to assess its pharmacokinetic properties and provide a basis for subsequent drug development and clinical trials.
  • Safety assessment. Safety assessment of the antibody/peptide, including acute toxicity test, long-term toxicity test, teratogenicity test, etc., in order to determine its toxic side effects and safe dose range.
  • Production process development. Development of efficient antibody/peptide production processes to ensure the quality and efficiency of the mass production process.

Multiple Immunization Strategies Based Antibody/Peptide Development

Membrane protein immunization. Typically involves extracting and purifying membrane proteins from cell membranes, which are then used to immunize animals or volunteers to generate antibodies or T-cell responses against specific membrane proteins. This immunization strategy helps to study the function of membrane proteins and also allows for the development of diagnostic reagents or therapeutic drugs for specific diseases.

Virus-like particles (VLPs) immunization. VLPs are nanoparticles that mimic the structure of viruses and are assembled from viral coat proteins or genetically encoded recombinant coat proteins. VLPs immunization focuses on stimulating organisms to produce antibodies against specific viruses by using VLPs as immunogens. This immunization strategy can be used to develop prophylactic vaccines and therapeutic antibodies against a wide range of viruses, such as human papillomavirus and hepatitis B virus.

DNA immunization. Direct injection of plasmid DNA encoding an antigen can induce an immune response against the antigen. This immunization strategy is highly effective, safe, and can induce long-lasting immunity. It can be used to rapidly assess the immunogenicity of antigens and to study the mechanism of the immune response to antigens in vivo.

Comprehensive Antibody/Peptide Screening Strategies

  • Immunoscreening. Utilizing the specificity of the antigen-antibody reaction, antibodies/peptides that bind to specific antigens are screened by immunological methods.
  • Affinity screening. Determine the affinity of the antibody/peptide to the antigen and screen for antibodies/peptides with higher affinity.
  • Functionality screening. Antibodies/peptides with specific functions are screened by measuring their functional activities, such as mediating immune responses, inhibiting pathogenic microorganisms, etc.
  • Cellular screening. Screening for antibodies/peptides that bind to cells or affect cellular functions by reacting with specific cell lines.
  • Transgenic or knockout screening. Screening for antibodies/peptides with specific gene expression by means of transgenesis or gene knockout using genetic engineering techniques.
  • Chemical screening. Chemical synthesis is used to synthesize a large number of different antibodies/peptides, and then specific screening methods, such as affinity chromatography and mass spectrometry, are used to screen out the target antibodies/peptides.
