Introduction of Peptide Immunogen

Peptide antigens are produced synthetically from carefully selected short amino acid sequences of target proteins. When target proteins are limited or unavailable, they are a popular solution because they are easy to synthesize and use with high purity. The peptide is covalently coupled to the carrier protein, and the resulting conjugate is used for immunization. In this case, the antibody produced is said to be sequence-specific. Such antibodies can cross-react with corresponding proteins and the epitopes of which can be easily mapped and identified. Using this method, antibodies can be generated against selected regions, such as highly conserved regions, active sites, extracellular or intracellular domains, or post-translational/chemically modified regions.

Peptide Selection

The choice of peptide immunogen depends on the intended use, i.e. the peptide antibody should recognize the putative antigen in denatured form (Western blotting) or natural form (immunoprecipitation or immunoassay). Especially if the target is a native protein, several questions need to be noted. The common feature of immunogenic peptides is that they preferentially represent sequential epitopes. For practical reasons, the selected amino acid sequence is usually determined by B-cell epitope examination or by processing physicochemical characteristics (e.g., accessibility, hydrophilicity, flexibility) and/or structural characteristics (e.g., loops, bends).
