General Materials

  • Enzymes and Solutions: Cell dissociation is performed using enzyme solutions, such as trypsin-EDTA, and washed with buffers.
  • Antibiotics and antimycotics: Prevent contamination risks.
  • Cryopreservation Media: Necessary for long-term preservation of stem cells.
  • Aseptic Equipment: Ensure aseptic conditions with sterile pipettes, petri dishes and centrifuge tubes.

Common Procedures

  • Preparation of Culture Medium
    Mix the medium with other required supplements to make a nutrient-rich medium. Maintain sterility throughout.
  • Cell Seeding and Maintenance
    Thaw frozen stem cells and transfer them to culture flasks containing culture medium and incubate them in an incubator.
  • Feeding and Passaging
    When cells reach 70-80% confluence, cells are separated by enzyme and collected by centrifugation. Resuspend the cells with fresh medium and transfer them to a new culture vessel. Maintain proper cell density for optimal growth.
  • Quality Control
    Perform regular viability and sterility tests to ensure the health of the cell culture. Verify cell identity through labeling and genetic analysis.
  • Differentiation and Characterization
    Use specific differentiation media to induce stem cell differentiation. Monitor cell morphology and expression of cell lineage-specific markers.
