The vector of anti-L1CAM chimeric antigen receptor (CAR) is constructed for the engineering of macrophage cells to target human L1CAM and induce target cell phagocytosis. The macrophage cells are genetically modified through transduction with a lentiviral vector expressing scFv of anti-L1CAM antibody (H2) linked to Megf10 signaling domains. The vector product was designed for the treatment of Ovarian cancer.

  • Megf10:
    Megf10 is used as the CAR intracellular signaling domain. As a phagocytosis receptor, Megf10 has cytosolic Immunoreceptor Tyrosine-based Activation Motifs (ITAMs) that are phosphorylated by Src family kinases. Based on this mechanism, Megf10 signaling can induce macrophage phagocytosis which has been demonstrated in the published article.
    It is a membrane receptor involved in phagocytosis by macrophages and astrocytes of apoptotic cells. It's a receptor for C1q, an eat-me signal, that binds phosphatidylserine expressed on the surface of apoptotic cells

  • Introduction
  • The protein encoded by this gene is an axonal glycoprotein belonging to the immunoglobulin supergene family. The ectodomain, consisting of several immunoglobulin-like domains and fibronectin-like repeats (type III), is linked via a single transmembrane sequence to a conserved cytoplasmic domain. This cell adhesion molecule plays an important role in nervous system development, including neuronal migration and differentiation. Mutations in the gene cause X-linked neurological syndromes known as CRASH (corpus callosum hypoplasia, retardation, aphasia, spastic paraplegia and hydrocephalus). Alternative splicing of this gene results in multiple transcript variants, some of which include an alternate exon that is considered to be specific to neurons.
