  • The optimized single-tube program can synthesize double-stranded cDNA up to 3 kb from total RNA or mRNA.

    The cDNA synthesis system is based on the method of Gubler and Hoffmann and uses an optimized one-tube procedure.

    Measurement time: 60 minutes for the first strand synthesis, 120 minutes for the second strand synthesis

  • 1. MV Reverse Transcriptase Buffer, 5x concentrated
    2. AMV Reverse Transcriptase (25 U/μl)
    3. 1,4-Dithiothreithol (0.1 M)
    4. Protector RNase Inhibitor (25 U/μl)
    5. Oligo (dT)15 primer, 200 μM (1 μg/μl)
    6. Oligo [(dT24 )T7promotor]65 primer, 100 μM (2 μg/μl)
    7. dNTP mixture (each nucleotide, 10 mM)
    8. Control neo mRNA (0.2 μg/μl)
    9. 2nd strand synthesis buffer, 5x concentrated
    10. 2nd strand enzyme blend (mixture of DNA Polymerase I, E. coli Ligase and RNase H)
    11. T4 DNA Polymerase (1 U/μl)
    12. Double-distilled water
    13. RNase I (10 U/μl)
    14. Proteinase K, recombinant
