Mass spectrometry imaging (MSI) is a technique that visualizes the detailed, sub-mm spatial distribution of analytes, thus enabling the exploration of target compounds at an unprecedented fine-scale and level of detail. This technique provides the opportunity to investigate tumor biology from an entirely novel biochemical perspective and could lead to the identification of a new pool of cancer biomarkers. Introduction of MSI Technology

MSI technology represents a highly promising approach for biomarker discovery in cancer research. Ion traps have been employed in biomarker discovery. Ion traps can store ions by applying multiple electrical fields from different directions. MALDI is one of the most frequently used ionization sources in protein biomarker discovery. The molecules ionized by MALDI are singly charged, which greatly simplifies the process of data analysis. As a widespread ionization technique employed in MSI, MALDI MSI emerges as a key technology for label-free mapping of the spatial distribution of biomolecules, ranging from small metabolites to large proteins. Combining all the advantages of modern mass spectrometry (MS), MSI achieves spatial resolutions allowing for cellular and, in some cases, even sub-cellular resolution. Therefore, as a rapidly maturing analytical technology, MSI is highly valuable for biomarker discovery.

MSI for Biomarker Discovery at Creative Biolabs

Many of MSI's features make it ideally suited to clinical research pathology applications. With years of experience in employing MALDI MSI for biomarker discovery, analytical sensitivity and specificity, spatial resolving capabilities, sample throughput, data handling, and statistical analyses have been greatly improved by scientists at Creative Biolabs. MSI allows measurement of small-sized samples and good specimen preservation for additional studies. What is more, it can use the formalin-fixed paraffin-embedded (FFPE) tissue sections. FFPE maintains excellent tissue morphology and allows the direct collection of both spatial and molecular information. Since the tissue acquisition method of FFPE doesn't require analyte-specific reagents, MSI is valuable for biomarker discovery. This technology can also be combined with other technologies, such as digital PCR, to make the best use of limited biopsy tissue. Except for the identification of protein biomarkers, MSI can also be used for the analysis of small-molecule metabolites, lipids, pharmaceutical compounds, and agrochemicals in plant tissues.
