Magnetic-activated cell sorting (MACS), also known as immunomagnetic cell sorting, is a well-established technology to separate desired cell populations depending upon cell surface antigens. Developed by Miltenyi in 1990, MACS uses antibodies, enzymes, lectins, or streptavidin bound to magnetic beads forming magnetic particles, which can specifically associate with surface markers or biotinylated molecules on the targeted cells. Thus, the target cells are labeled by the magnetic beads. Then, the cell suspension is transferred to a column or placed in an external magnetic field, the magnetically labeled target cells are magnetized to the walls of the column or polarized, whereas nonlabeled cells are washed away. The bead-captured cells are released and collected for recovery when the magnetic field is turned off. MACS can be divided into column-based magnetic cell separation and column-free magnetic cell separation according to the separation method, and positive or negative isolation according to the separation mode.
MACS is a comparatively simple, high-throughput, and cost-effective technology. MACS has been commonly used in the application areas of immunology, cancer research, neuroscience, and stem cell biology due to its high purity, recovery, and throughput.

Efficiently isolating antigen-specific antibody-secreting B cell populations from heterogeneous mixtures is a key part of rapid Native™ antibody development. MACS-based antigen-coated magnetic beads sorting is preferred to high throughput isolate antigen-specific B cells or plasma cells efficiently.
Distinct from general immunomagnetic cell sorting that uses magnetically labeled antibodies to localize target cell antigens, the improved antigen-coated magnetic beads sorting uses antigen-coated magnetic beads to isolate target antibody-secreting B cells. Antigen-coated magnetic beads can recognize and specifically bind to membrane anti-antigen antibodies on the B cell surface, by which those B cells coated by antigen magnetic beads are magnetized and trapped in the field when exposed to an external magnetic field. B cells and other cells, which do not secrete specific antibodies, do not bind to magnetic beads and are easily washed away. The target B cells are obtained by removing the external magnetic field. This antigen-coated magnetic beads sorting can separate a large number of high purity antigen-specific B cells from complex samples in a few minutes.
