Why Focus on Canine Parvovirus?
Canine Parvovirus is a highly contagious, non-enveloped DNA virus that primarily targets rapidly dividing cells in the intestinal crypts and bone marrow. It leads to severe hemorrhagic gastroenteritis, immunosuppression, and high mortality in young dogs. Despite the availability of vaccines, maternal antibody interference and viral antigenic variation can compromise protective immunity. Therapeutic antibodies targeting CPV capsid proteins offer immediate passive immunity, capable of neutralizing circulating virus and preventing systemic infection. Such biologics can serve as emergency treatments or as adjuncts in outbreak management, especially for unvaccinated or high-risk dogs.
Mechanism of Action
CPV-targeting antibodies function primarily through virus neutralization, binding to epitopes on the viral capsid proteins (VP1/VP2) and preventing viral attachment to the canine transferrin receptor (TfR), which is required for host cell entry. By blocking this critical step, the antibodies inhibit viral internalization and replication in the intestinal and hematopoietic tissues. Additionally, Fc-mediated effector functions, such as antibody-dependent cellular cytotoxicity (ADCC) and complement activation, contribute to viral clearance. This dual mechanism enables therapeutic antibodies to both prevent infection and mitigate disease severity when administered post-exposure.
