CD33 (Siglec-3) is a transmembrane receptor expressed primarily on myeloid lineage cells, such as monocytes, macrophages, and some dendritic cells. Functionally, CD33 acts as an inhibitory receptor, modulating immune activation through ITIM-mediated signaling to maintain homeostasis and prevent excessive inflammation. In dogs, aberrant CD33 signaling has been linked to chronic inflammatory diseases and immune cell dysfunction. In cats, research suggests potential relevance to myeloproliferative and immune-mediated disorders. By regulating myeloid cell activation, CD33 presents an attractive therapeutic target for managing both autoimmune and inflammatory diseases in pets, and potentially for oncology indications where immune modulation plays a central role. Mechanism of Action

Therapeutic antibodies targeting CD33 function by modulating its inhibitory signaling pathway on immune cells. Upon antibody binding, CD33 signaling can be altered, either suppressed or blocked, affecting downstream phosphorylation of ITIM motifs and subsequent recruitment of phosphatases like SHP-1 and SHP-2. This modulation can restore immune balance by reducing hyperactivation in inflammatory diseases or by enhancing immune cell activity in oncology contexts. In companion animals, CD33-directed antibodies may therefore serve dual functions: immunosuppression in chronic inflammatory states or immune activation when combined with immunostimulatory agents.
