Why Focus on IL-31?
IL-31 is a pro-inflammatory cytokine produced predominantly by activated T cells and plays a central role in mediating pruritus (itching) and skin inflammation. In dogs, IL-31 is strongly associated with atopic dermatitis, one of the most prevalent chronic skin diseases in veterinary medicine. This condition leads to persistent itching, self-trauma, secondary infections, and reduced quality of life. While IL-31's role in feline dermatologic disorders is less studied, emerging evidence suggests it may contribute to allergic skin disease in cats as well. Given the significant burden of chronic pruritus in companion animals, targeting IL-31 presents a powerful therapeutic opportunity to improve comfort and long-term disease management.
Mechanism of Action
IL-31 antibodies work by binding to circulating IL-31 and preventing its interaction with the IL-31 receptor complex, which is expressed on keratinocytes, sensory neurons, and immune cells. By blocking this ligand-receptor interaction, antibody therapy interrupts downstream signaling pathways that drive itch sensation and skin inflammation. This not only reduces scratching behavior but also improves skin barrier integrity and lowers the risk of secondary bacterial or yeast infections.
