Why Focus on IL-5 / IL-5R?
IL-5 is a critical cytokine involved in eosinophil activation, survival, and recruitment, while IL-5R serves as its signaling receptor. Dysregulation of this pathway is linked to eosinophilic disorders, allergic inflammation, and asthma-like conditions in companion animals. In dogs, eosinophilic bronchopneumopathy is a notable example of IL-5–driven pathology, while in cats, eosinophilic granuloma complex and allergic airway disease present similar immunological challenges. By targeting IL-5 or IL-5R, therapeutic antibodies offer a promising strategy to reduce excessive eosinophil activity, alleviate inflammation, and improve long-term outcomes for affected pets.
Mechanism of Action
Therapeutic antibodies against IL-5 or IL-5R work by disrupting the signaling cascade responsible for eosinophil proliferation and survival. Anti-IL-5 antibodies neutralize the cytokine, preventing its interaction with the receptor, while anti-IL-5R antibodies directly block receptor activation. Both approaches reduce circulating and tissue-resident eosinophil counts, mitigating inflammation and tissue damage. In pets, this translates into reduced respiratory distress, improved skin health, and better management of chronic allergic conditions, providing a targeted alternative to corticosteroids and other non-specific treatments.
