Why Focus on TNF-α?
TNF-α is a pro-inflammatory cytokine that plays a critical role in mediating immune responses. While essential for host defense, excessive or dysregulated TNF-α activity contributes to chronic inflammatory diseases, autoimmune disorders, and certain cancers in pets. In dogs, elevated TNF-α levels are implicated in conditions such as inflammatory bowel disease (IBD), rheumatoid arthritis, and atopic dermatitis. In cats, TNF-α is linked to feline chronic gingivostomatitis and other inflammatory disorders. By targeting TNF-α, therapeutic antibodies can modulate pathological inflammation, reducing tissue damage and improving clinical outcomes. This makes TNF-α a highly relevant and actionable target for both canine and feline patients, addressing conditions that currently have limited treatment options.
Mechanism of Action
TNF-α-targeting antibodies function by binding to circulating TNF-α molecules, preventing their interaction with TNF receptors (TNFR1 and TNFR2) on target cells. This blockade inhibits downstream pro-inflammatory signaling pathways, including NF-κB activation, which is responsible for driving cytokine production, immune cell recruitment, and tissue damage. By neutralizing TNF-α activity, these antibodies can effectively reduce inflammation, alleviate clinical symptoms, and halt the progression of immune-mediated diseases in dogs and cats. Depending on the disease context, this mechanism can provide both systemic and localized therapeutic benefits.
