Previously, it is difficult to isolate a purified glycosyltransferase with enough quantity for composition, structure and function analysis because the majority of glycosyltransferases are in low abundance. Expression cloning glycosyltransferase genes is a method to amplify the production of desired glycosyltransferases by expressing encoding cDNA, which offers great convenience for characterizing and analyzing glycosyltransferases. The advent of this technique was originally dated back to 1987 by Seed and Aruffo, which has ushered the research and development of glycosyltransferases into a new era. Expression cloning glycosyltransferase makes it possible to clone a glycosyltransferase encoding cDNA without the isolation and purification, that recognizes a specific carbohydrate by specific reagents. The roles of an increasing number of carbohydrates have been elucidated based on these newly cloned glycosyltransferases, which undoubtedly provide us with a better understanding of biological functions and the importance of carbohydrates.
Carbohydrates, as known to all are one of the major nutrients, are non-primary products of gene expression that synthesized by glycosyltransferases. Carbohydrates are critical bioactive compounds and energy provider of human beings, but they are also possible to be antigens with the ability to induce antibody production without the help of T lymphocytes. Anti-glycan antibodies are a group of antibodies that directly target carbohydrate antigens, which involve numerous biological processes and widely used in glycobiology.
Anti-glycan antibodies are one of the most widely utilized reagents in expression cloning glycosyltransferase based on the specificity and high affinity. An approach using anti-glycan antibodies for expression cloning glycosyltransferases is similar to other antibodies, of which glycosyltransferases are enriched by transfected cell amplification. Selection of the cells transfected with a glycosyltransferases cDNA expression library by using an anti-glycan antibody specific reaction to the desired cloned antigen, afterward, scanning and amplifying these selected cells to enlarge the expression of glycosyltransferases.
